Kisspeptin-10 is a truncated, biologically active fragment of the kisspeptin family of peptides, derived from the KiSS-1 gene product. It is the most upstream regulator of the hypothalamic-pituitary-gonadal (HPG) axis, directly stimulating GnRH neurons to initiate the reproductive hormone cascade. It is an emerging research compound in reproductive endocrinology, with applications in fertility diagnostics, IVF protocols, and understanding puberty initiation.

Mechanism of Action

C-terminal decapeptide fragment of kisspeptin that activates GPR54 (KISS1R) receptors on GnRH neurons. Potently stimulates the hypothalamic-pituitary-gonadal (HPG) axis, triggering pulsatile GnRH release and downstream LH and FSH secretion.

Clinical Research Status

Kisspeptin-10 is in early-phase clinical research primarily at academic centers, with Imperial College London leading human studies for IVF and reproductive disorders. Clinical trials have demonstrated its ability to trigger LH pulses and stimulate oocyte maturation as a potential alternative to HCG triggers in IVF. It is not yet approved for any therapeutic indication but has orphan-like research status in reproductive medicine.

Key Published Findings

Human studies demonstrate that kisspeptin-10 infusion (at doses of 0.1-1.0 nmol/kg) potently stimulates LH and FSH secretion in healthy volunteers and patients with reproductive disorders. In IVF contexts, kisspeptin-triggered oocyte maturation has produced viable pregnancies with theoretically lower risk of ovarian hyperstimulation syndrome compared to HCG triggers. Research confirms kisspeptin neurons are the critical gatekeepers of puberty onset, integrating metabolic and circadian signals to initiate HPG axis activation.

Safety Profile

Kisspeptin-10 has demonstrated an excellent safety profile in human clinical studies, with no serious adverse events reported across multiple Phase 1/2 trials. Its short half-life (approximately 4-5 minutes for kisspeptin-10) means effects are rapidly reversible, reducing risk of prolonged overstimulation. Unlike HCG triggers in IVF, kisspeptin stimulation produces a more physiological gonadotropin surge that may reduce ovarian hyperstimulation syndrome risk.

Comparison to Related Compounds

Unlike gonadorelin which directly activates GnRH receptors on the pituitary, kisspeptin acts one step further upstream by stimulating GnRH neurons themselves, producing a more physiological hormonal cascade. Compared to HCG for IVF triggering, kisspeptin produces a shorter and more physiological LH surge. The kisspeptin-54 (full-length) form has a longer half-life than kisspeptin-10 but both activate the same GPR54 (KISS1R) receptor.

Community Observations

Kisspeptin-10 has attracted interest from reproductive endocrinology researchers as a potentially safer alternative for IVF trigger protocols, particularly in OHSS-prone patients. The peptide's position at the apex of the HPG axis makes it a uniquely informative diagnostic tool for distinguishing hypothalamic from pituitary causes of hypogonadism. Its extremely short half-life currently limits practical therapeutic applications outside of controlled clinical settings.
